Structura tezei
Prezenta teză de doctorat cu titlul „Materiale biodegradabile cu antioxidanti naturali
pentru protectia mediului si aplicatii biomedicale” este alcătuită din 4 capitole, ce însumează
un studiul amplu al literaturii ştiinţifice de actualitate, dar şi o partea experimentală, caracterizată
de o multitudine de date privind stabilizarea materialelor biodegradabile cu antioxidanţi naturali.
CAPITOLUL 1 face referire la poziţionarea epistemologică a cercetării, precizându-se
obiectivul şi problematica cercetării, ipotezele, metode analitce şi statistice de cercetare şi
structura tezei.
CAPITOLUL 2 include partea teoretică cu privire la stabilizarea materialelor
biodegradabile cu antioxidanţi naturali, explicându-se proprietăţile materialelor polimerice
studiate, dar şi a antioxidanţilor naturali. În acest capitol sunt aboradate problemele actuale ale
cercetării privind biodegradabilitatea materialelor utilizate şi perspectivele acestora.
CAPITOLUL 3 poate fi numit cu siguranţă cea mai preţioasă şi importantă parte a acestui
studiu. În acestă parte a tezei au fost caracterizate materialele folosite, tehnicile analitice cu
ajutorul cărora s-au făcut investigaţiile asupra materialelor, metodele statistice folosite pentru
interpretarea rezultatelor.
Antioxidanţii naturali (şi extractele cu conţinut ridicat al acestora) au fost caracterizaţi
prin spectrofotometria UV-Vis, folosind metoda Folin-Ciocalteu (la aproximativ 760 nm), dar şi
prin metoda determinării conţinutului de compuşi fenolici la 280 nm. Au fost efectuate studii
privind stabilitatea resveratrolului (antioxidant natural) la temperatură ridicată cu ajutorul
spectrofotometriei UV-Vis şi spectroscopiei FT-IR.
Extractele din pieliţă de struguri au fost analizate prin spectrofotometria UV-Vis, folosind
metoda Folin-Ciocalteu, dar şi prin spectroscopia FT-IR şi calorimetria diferenţială (DSC).
Studiile pe gelatină au fost efectuate folosind spectrofotometria UV-Vis şi spectroscopia
FT-IR.
Pentrul studiul polistirenului stabilizat cu material polifenolic (extract din pieliţă de
struguri) au fost folosite calorimetria diferenţială (DSC), spectroscopia FT-IR, microscopia
electronică cu baleiaj (SEM) şi cromatografia ionică (IC).
Studiile legate de chitosan, alcool polivinilic, colagen şi hidroxilat de calogen au fost
efectuate prin spectroscopia FT-IR.
CAPITOLUL 4 atinge concluziile principale privind acestă lucrare ştiinţifică, sublinind
elementele de originalite şi de noutate din această teză.
CAPITOLUL 5 cuprinde referintele bibliografice utilizate în cadrul acestei teze de
doctorat.
CAPITOLUL 1. DEMERSUL EPISTEMOLOGIC AL CERCETĂRII
Pe fondul epuizării rezervelor de petrol ale planetei, cercetările actuale sunt orientate spre
utilizarea materiilor prime regenerabile şi producerea de materiale biodegradabile. Din
această perspectivă, materiile prime vegetale şi în mod special biopolimerii, posedă proprietăţi de
mare interes în industria de materiale plastice cum ar fi: biodegradabilitatea, biocompatibilitatea,
permeabilitatea selectivă şi proprietăţile fizico-mecanice modificabile.
Posibilitatea obţinerii de materiale noi, cu proprietăţi îmbunătăţite, prin amestecarea a doi
sau mai mulţi polimeri reprezintă o soluţie practică mai bună, din punct de vedere economic,
decât proiectarea şi sinteza de noi polimeri. În ultimele decenii, cercetările ştiinţifice au fost
orientate către dezvoltarea unei noi clase de polimeri, cea a biomaterialelor.
Unul din cei mai discutaţi antioxidanţi naturali rămâne resveratrolul. Proprietăţile
acestuia au fost foarte intens studiate de cercetători, care au identificat mai ales efecte
benefice asupra sănătăţii [1; 2].
Pentru a putea integra resveratrolul în structura diverselor biomateriale, au fost
necesare studii privind modul de introducere a acestuia în matricea materialelor
biodegradabile. Cel mai important aspect este stabilitatea resveratrolului la temperatură,
pentru că majoritatatea materialelor polimerice sunt prelucrate prin tratament termic.
Există şi alte modalităţi de integrare a antioxidanţilor în structura materialelor
biodegradabile. Pe baza solubilităţii polimerului într-un anumit solvent este posibilă
introducerea anumitor antioxidanţi în polimer. De exemplu, gelatina poate fi îmbogăţită cu
antioxidant prin dizolvare în apă, iar polistirenul – prin dizolvare în cloroform.
În prezent, interes deosebit prezintă polimerii biodegradabili, precum: chitosanul,
colagenul, hidroxilatul de colagen şi alcoolul polivinilic. Aceştia sunt de multă vreme
utilizaţi în aplicaţiile biomedicale şi au grad înalt de biocompatibilitate. Prin stabilizarea
acestor polimeri cu antioxidanţi naturali, poate creşte durata de viaţă a acestor materiale,
dar scade şi riscul de intoxicare datorită înlocuirii antioxidanţilor artificiali cu cei naturali.
La general vorbind, interacțiunea dintre orice material polimeric cu antioxidanţi
naturali aduce un avantaj considerabil privind biodegrabilitatea pentru mediu. În domeniul
medical, folosirea antioxidanţilor naturali în structura diferitor materiale biodegradabile, ar
produce numeroase efecte pozitive.
Obiectivul principal al acestei teze a fost de a produce materiale biodegradabile
stabilizate cu antioxidanţi naturali cu aplicaţii în medicină şi pentru protecţia mediului.
Problematica cercetării a plecat de la toxicitatea produsă de antioxidanţii artificiali asupra
mediului. De asemenea, în cazul unor materiale polimerice greu degradabile înlocuirea
antioxidanţilor artificiali cu cei naturali ar micşora durata de degradare a acestora şi ar creşte
şansele pentru biodegradare.
CAPITOLUL 2. DEGRADABILITATEA ŞI BIODEGRADABILITATEA
MATERIALELOR POLIMERICE ÎN CONDIŢII DE MEDIU
Polimerii de sinteză au generat un mare entuziasm din punct de vedere tehnologic, astfel
că la mijlocul anilor ‘50 punerea în funcțiune de noi instalații de producție și cercetarea obținerii
de noi structuri polimerice pentru aplicații civile, industriale și militare erau în plin avânt.
Conștientizarea problemelor pe care aceste materiale le pot crea asupra mediului a apărut mult
mai târziu, atunci când acumularea și persistenţa acestora la gropile de gunoi sau în mediul
ambient a devenit evidentă, când efectele asupra florei, faunei și asupra calităţii solului și calităţii
apei au fost dovedite și înțelese într-o măsură mai mare. În plus, criza petrolului de la sfârşitul
anilor ’70 a relevat faptul că asigurarea resurselor de materii prime pentru continuarea şi
dezvoltarea producţiei de materiale polimerice ar putea fi ea însăşi o problemă în viitor [3].
În condiţiile utilizării unor materiale provenite din resurse regenerabile, care ar contribui
la menţinerea practic constantă a concentraţiei CO2 din atmosferă, atât eliminarea deşeurilor de
materiale plastice prin combustie cât și prin degradare în mediul înconjurător pot câştiga în
eficienţă, datorită utilizării unor surse de materii prime alternative celor fosile. Un studiu
intermediar îl reprezintă utilizarea unor materiale polimerice tradiţionale ale căror proprietăţi
sunt controlate prin intermediul unor aditivi ce au un impact mai mic asupra mediului şi sănătăţii
umane. Astfel, este cunoscută utilizarea antioxidanţilor fenolici de sinteză pentru stabilizarea
poliolefinelor în cursul procesării şi pentru asigurarea unei anumite durabilităţi în timpul
utilizării. Este cunoscut că antioxidanţii fenolici pot prezenta activitate bactericidă importantă şi
chiar o anumită toxicitate pentru animale şi om. Aceşti aditivi pot migra în alimentele aflate în
contact cu materialele polimerice de ambalaj, dar şi în sol sau în apă în condiţiile degradării
materialului polimeric în mediul ambiant. [4]
O soluţie de reducere a riscurilor legate de utilizarea aditivilor (antioxidanţi, stabilizatori)
asupra sănătăţii umane şi a mediului înconjurător o reprezintă utilizarea unor produse naturale
sau a unor produşi de sinteză cu structură similară celor naturale [4].
Preocupările privind găsirea unor structuri polimerice capabile să sufere degradare în
medii înconjurătoare active (Figura 1), sub acţiunea microorganismelor, au început în anii '80.
Materialele plastice biodegradabile au deschis astfel drumul pentru noi strategii în managementul
deşeurilor deoarece astfel de materiale sunt concepute să se degradeze în condiţiile mediului
înconjurător sau în instalaţii industriale de tratare biologică a deşeurilor [5].

Evaluarea susceptibilităţii la biodegradare şi stabilirea mecanismelor de reacţie, a
factorilor de influenţă şi a ponderii acestora oferă atât posibilitatea dirijării procesului de
biodegradare, cât şi a sintezei de materiale ale căror deşeuri să se integreze facil în ecosistemele
naturale. [7]
După expunerea în condiţii definite prin standarde internaţionale (ASTM – American
Standard Testing Methods sau ISO – International Standards Organization), probele sunt
analizate din punct de vedere al proprietăţilor fizice, chimice sau biologice. Rezultatele obţinute
se compară cu valorile omonime obţinute la probele de control.
Astfel, s-au pus la punct teste de biodegradare în sol sau în nămol activ în condiţii
controlate de temperatură, umiditate şi aerare. În general, testarea biodegradabilităţii poate fi
efectuată conform protocolului din figura 2. [8]

În prezent se fac eforturi pentru transforma polimerii nedegradabili existenţi în polimeri
biodegradabili, fotodegradabili sau hidrolizabili prin modificarea chimică sau prin includerea de
aditivi (de exemplu, sensibilizatori şi biopolimeri) şi prin dezvoltarea unor metode care fac
posibilă utilizarea de polimeri biodegradabili nativi [de exemplu, poli (β-hidroxibutiratul)]. [9;
10]
Utilizarea adecvată a polimerilor biodegradabili în diferite aplicații depinde de
cunoaşterea interacţiunii dintre aceste materiale și mediul înconjurător. Această interacţiune este
controlată, între altele, de tipul şi cantitatea moleculelor mici care pot penetra materialul sau se
degajă din acesta prin difuzie şi migraţie [10]. Obținerea industrială a multora dintre polimerii
biodegradabili este încă foarte costisitoare pentru aplicații cum sunt articolele de unică utilizare

(filme de ambalare, scutece, tacâmuri de unică folosinţă), dar într-o serie de aplicaţii
biomedicale, costul are o importanţă minoră.
În prezent, antioxidanţii sunt utilizaţi pe scară largă în industria alimentară pentru a
preveni sau întârzia oxidarea lipidelor. Controlul oxidării este esențial în gestionarea evoluţiei
sistemelor biologice în complexitatea lor, în special în cazul produselor alimentare deoarece
degradarea poate afecta siguranța alimentelor (Figura 3). [11]
Există mai multe criterii care trebuiesc luate în considerare atunci când se aleg
antioxidanţii corespunzători pentru a fi incluşi în produsele alimentare, cum ar fi: preţul de cost,
toxicitatea acestuia, eficacitatea lui la concentraţii mici, stabilitatea şi capacitatea de procesare,
culoarea, aroma, şi mirosul [12].

Rolul compuşilor fenolici în organism este în general de protecţie împotriva anumitor
boli datorită posibilelor interacţiuni cu enzime şi proprietățile lor antioxidante. În mod specific,
flavonoidelor le sunt atribuite o mare varietate de proprietăți, şi anume: antitumorale,
antiinflamatoare, analgezice, antialergice, antispastice, antibacteriene, hepatoprotectoare [13].
Acestea limitează oxidarea grăsimilor din sânge şi contribuie la lupta împotriva plăcilor
ateromatoase. De asemenea au proprietăţi anxiolitice protejând arterele împotriva aterosclerozei
şi a trombozei. [14]
CAPITOLUL 3. PARTEA EXPERIMENTALĂ
3.1. Modul de pregătire a probelor
În cadrul lucrării am folosit probe de extracte de struguri, aparţinând mai multor soiuri şi
anume: Căpşunică, Tămâioasă neagră, Hamburg, Moldova, Coarne Moldoveneşti (notate cu P1,
P2, P3, P4, P5).
Am ales etanolul ca solvent de extracţie deoarece nu este toxic, fiind indicat în numeroase
studii şi datorită capacităţii de a izola un spectru mai larg de compuşi apolari şi polari din probă.
Pieliţele de struguri au fost îndepărtate manual, apoi cântărite (~ 100 g pieliţă) şi introduse în 50
ml etanol 96%. După omogenizare, eşantioanele au fost ţinute la întuneric timp de 48 de ore.
Extractele au fost apoi obţinute prin centrifugare la 4000 rpm timp de 20 minute, au fost filtrate
prin filtre cu diam. 0,45μm şi păstrate la întuneric.
Prepararea soluţiilor apoase de trans-resveratrol
Trans-resveratrolul (substanţă pur analitic 98%) se diluează în 200ml H2O, rezultând o
soluţie de 31.25mg/L (137μM). Din această soluţie s-au obţinut soluţii apoase de trans-
resveratrol cu concentraţii de 5, 10, 15.625, 25 şi respectiv 31.25mg/L.

Pentru a determina concentraţia resveratrolului în extractele studiate, s-au efectuat 3
curbe de calibrare la 3 lungimi de undă diferite (252, 286, 304 nm) [15] folosind resveratrol pur
analitic (Merck) şi utilizând un spectrofotometru Analityk Jena- Specord 250 Plus.
Concentraţia de echivalent-resveratrol din extractele de struguri a fost determinată
folosind curba de calibrare a resveratrolului la 304 nm (Figura 4).

Datele preliminare de spectroscopie UV-VIS au arătat un conţinut de echivalent
resveratrol mai mare în eşantioanele P1 şi P4 şi mai mic în P2 şi P3.
3.2. Determinarea indicelui total de fenol din extractele de struguri
Indicele total de polifenoli s-a determinat prin măsurarea absorbanţei la 280 nm utilizând
cuve cu drum optic de 10 mm şi făcând diluţie de 1:100 (Figura 5) [16, 17]. Rezultatele obţinute
sunt în bună corelaţie cu concentraţia de echivalent-resveratrol din extractele de struguri. Astfel,
conţinutul de polifenoli este mai ridicat în eşantioanele P1 şi P4, şi mai scăzut în eşantioanele P2
şi P3. Un fenomen foarte interesant poate fi observat între indicele total de polifenoli şi
concentraţia de echivalent-resveratrol din extracte (Figura 6). Putem observa o relaţie aproape

liniară între acestea. Desigur, relaţia matematică între aceşti doi indici (R2
= 0,8576) poate fi
rezultatul apropierii între lungimile de undă la care s-au efectuat măsurătorile (280 şi 304 nm).

3.3. Determinările FT-IR efectuate pe extractele de struguri şi resveratrol

Cu ajutorul acestei tehnici s-au identificat grupările funcţionale ce intră în compoziţia
extractelor de struguri (Figura 7) şi s-au obţinut mai multe date privind extractele analizate.
Pentru înregistrarea spectrului s-a utilizat o soluţie apoasă de trans-resveratrol având concentraţia
de 15,625 mg/L.

Din analiza graficelor obţinute se pot observa peak-urile 1240 cm-1
, 600 – 800 cm-1
, 3200
– 3400 cm-1
, picuri care caracterizează grupări funcţionale prezente în compuşi aromatici. După
cum se cunoaşte, în struguri cei mai întâlniţi compuşi aromatici sunt polifenolii. Se poate
concluziona analizând informaţiile obţinute că spectroscopia FT-IR poate fi utilă la detectarea
polifenolilor (figura 7-8).
3.4. Determinări de calorimetrie diferențială efectuate pe extractele de struguri
Măsurătorile legate de calorimetrie diferenţială (DSC) au fost efectuate pe 2 extracte de
struguri şi anume Căpșunică (notat cu Q1, Figura 9) şi Hamburg (notat cu Q2, Figura 10), care
au trecut înainte de aceasta printr-o pregătire specială (deshidratare la 400 °C pe perioade
îndelungate).

S-a observat că procesele care pot influența concentrația de polifenoli încep după
temperatura de 170oC, fiind vorba de oxidare, iar după 190
oC încep practic procesele de
descompunere. Este de remarcat faptul că la extractul Q1 aceste procese încep la temperaturi mai
ridicate (oxidare la 190oC, descompunere la 200
oC).
3.5. Caracterizarea activității antioxidante a extractelor de struguri prin metoda
Folin-Ciocalteu
S-a utilizat indicele Folin-Ciocalteu pentru a caracteriza activitatea antioxidantă a
extractelor de struguri, măsurătorile fiind efectuate la 4 lungimi de undă: 725, 750, 760 și 765
nm.
Corelaţiile de tip Pearson dintre valorile concentraţiei extractelor de struguri obţinute pe
curbele de calibrare la 4 lungimi de undă (725, 750, 760 şi 765 nm) au arătat asemănări foarte
mari (1,000, p < 0,01) (tabelul 1). Prin această analiză statistică nu s-au putut evidenţia
diferenţieri între concentraţiile de acid galic din probe.

S-au aplicat parametrii de probabilitate a concentrației reale de compuși fenolici în
extractele analizate. Prin analiza componentelor principale, urmată de metoda de rotație Varimax
cu normalizare Kaiser, s-au obținut 2 componente principale (Figura 11):
- componenta 1, caracterizată de valorile concentrațiilor extractelor de struguri, care
au prezentat abatere standard relativ mare;
- componenta 2, caracterizată de valorile concentrațiilor extractelor de struguri,
care au prezentat abatere standard relativ mică.

3.6. Investigaţii privind stabilitatea termică a resveratrolului
Eşantioane diferite de soluţie apoasă de trans-resveratrol (19 mg/L resveratrol) au fost
expuse la temperaturi de 30, 40, 50, 60, 70, 100 şi 120oC timp de 30 minute. De asemenea, alte
eşantioane având aceeaşi concentraţie au fost expuse la temperaturi de 30, 40, 50, 60, 70 oC
pentru 1200 minute (figura 12).
Pentru determinările privind stabilitatea trans-resveratrolului la temperatură s-a folosit o
soluţie de resveratrol de 19 mg/L. Investigaţiile efectuate au demonstat că temperatura are un
efect important asupra absorbanţei trans-resveratrolului la lungimile de undă 286 şi 304 nm.
Scăderea abosrbanţei demonstrează faptul că la temperaturi ridicate, resveratrolul se distruge. Nu
se poate admite nici conversia trans-resveratrolului în cis-resveratrol datorită faptului că şi cis-
resveratrolul prezent în soluţie pare să fie degradat.

Lungimile de undă la care s-au făcut determinările indicelui Folin-Ciocalteu sunt: 725,
750, 760 şi 765 nm (Figura 13). Standardele de calibrare au fost preparate dintr-o soluţie mamă
de 5 g/L acid galic.
Analizând spectrele soluțiilor de acid galic și aplicând cele 3 tipuri de analiză statistică
(regresie liniară, analiza componentelor principale, corelaţiile de tip Pearson) se poate
concluziona că lungimea de undă 725 nm este cea mai potrivită.
Astfel, la lungimea de undă de 725 nm există cea mai puternică corelație între valoarea
absorbanţei și concentrație, iar maximul de absorbţie se află în jur de 765 nm.
3.8. Studii privind interacţiunea sistemului gelatină-resveratrol-apă
Pentru a investiga comportamentul sistemului gelatină-resveratrol-apă s-au preparat 5
probe cu aceeași cantitate de gelatină, dar cu concentrații diferite de resveratrol. Probele
preparate au fost supuse unui tratament termic la temperaturile de 40, 60, 80 °C, iar apoi au fost
analizate prin spectrometrie FT-IR.
S-a observat că procesele de deshidratare pentru sistemul gelatină-resveratrol-apă nu sunt
afectate de cantitatea de resveratrol adăugată, iar probele se comportă asemănător la temperatura
de 40 °C (≥ 0,897, p < 0,01). Prin aplicarea corelațiilor Pearson s-a observat că rasveratrolul nu
are nici o influență sau are o influență nesemnificativă asupra pierderilor de masă de către probe.
Din analiza spectrelor FT-IR s-a observat că există diferențe între probe probe în domeniul 8000
- 400 cm-1
. Acest lucru poate fi datorat proteinelor din gelatină care hidrolizează în mod diferit,
deoarece s-a demonstrat si prin analiza statistica a componentelor principale ca la această
temperatură resveratrolul are o influență nesemnificativă asupra gelatinei.
Referitor la influența timpului asupra probelor de gelatină stabilizate cu resveratrol și
menținute la 60 °C, se observă că acesta ar avea un impact mai mic (≥ 0,601) comparativ cu
probele care au fost menținute la temperatura de 40 °C. Explicația poate fi în faptul că
temperatura mai ridicată accelerează pierderea de masă (mai ales pierderea apei) într-un timp
mai scurt. Între pierderile de masă a probelor se observă corelaţii foarte strânse (1,000; p < 0,01),
acestea demonstrând un mod echilibrat de transformare. Există o influență a resveratrolului
asupra sistemului observată după 48,06 h la temperatura de 60°C, dar caracterizată de o corelație
medie (0,487).
Proba P5 (cu cea mai mare concentrație de resveratrol) a prezentat un caracter
intermediar privind asemănarea dintre spectre. Distribuția probelor în grafic, din acest punct de
vedere, poate fi legată, după cum s-a mai arătat, de procesele de hidroliză și de refacerea
legăturilor la deshidratare, care au decurs într-un mod aleatoriu.
În ceea ce privește tratamentul termic la la temperatura de 80 °C, influența timpului este
descrisă de corelații destul de puternice (≥ 0,705). Sunt stabilite corelații foarte puternice între
valorile pierderilor de masă (≥ 0,988, p < 0,05), acest lucru demonstrând că deshidratarea
decurge în același mod la toate probele. Ca și în cazul celorlalte studii (la 40 și 60 °C), am
observat că timpul are influență asupra probelor, dar nu majoritară.
Din analizarea spectrelor și prin aplicarea metodelor de analiză statistică s-a observat că
probele P3_40, P4_40 şi P1_80 au transmitanţă mai ridicată în domeniul 3300 - 1082 cm-1
.
3.9. Studii privind comportamentul gelatinei stabilizate în diferite medii biologice
În prezenta lucrare a fost studiată interacţiunea dintre resveratrol şi gelatină, acid ascorbic
şi gelatină, extract struguri şi gelatină, extract rozmarin şi gelatină. Este foarte importantă
compatibilitatea dintre cele 2 componente, dar şi comportamentul fiecărui sistem in interacție cu
diverse fluide biologice: suc gastric, suc intestinal, plasmă sanguină.
Testul Folin-Ciocalteu a oferit o determinare foarte bună privind cantitatea de principii
active migrată din material în fluidele biologice.
Extractul de rozmarin a prezentat o hidrofobicitate ridicată în toate fluidele biologice
studiate, dar este posibil ca în mediul biologic natural lucrurile să decurgă altfel.
S-a observat, în toate fluidele analizate, după un interval de 96 h, o migraţie a principiului
activ (cuantificată în mg/L echivalent acid galic), iar cele mai mari valori au fost observate la
probele care au avut contact direct cu sucul gastric şi cu sucul intestinal. În concluzie, extractul
de rozmarin poate fi folosit ca si component în compoziţia principiilor uleioase sau în mediile în
care este nevoie ca antioxidantul să nu migreze (Figura 14).
În ceea ce privește cantitatea de extract de pieliţă de struguri (soi Căpşunică) adăugată în
probe, se observă că aceasta este direct proporţională cu concentraţia de echivalent de acid galic
la probele care au fost menţinute în suc gastric şi suc intestinal. Probele menţinute în plasmă
sanguină au demonstrat o creştere a compuşilor fenolici în raport cu timpul, dar necorelată cu
cantitatea de extract de struguri adăugată.

În sucul intestinal a avut loc cea mai uniformă eliberare a compuşilor polifenolici.
Probele de gelatină menţinute în suc gastric au avut un caracter intermediar, dar, care combinat
cu acţiunea sucului gastric ar putea accelera migraţia compuşilor polifenolici şi hidroliza
gelatinei.
Din figura 15 se observă că în toate fluidele biologice are loc difuzia. Totuşi în sucul
gastric şi în cel intestinal aceasta decurge într-un ritm mai accelerat.
În ceea ce privește acidul ascorbic s-a observat că migrația acestuia în gelatină este direct
proporţională şi cu durata contactului cu fluidele biologice, dar şi cu concentraţia de acid
ascorbic din probe. Cele mai mari valori ale concentraţiei de acid ascorbic se observă în sucul
gastric şi în sucul intestinal. Maximul de concentraţie înregistrat pe gelatina menţinută timp de
96 h în plasmă sanguină este de 2 ori mai mic faţă de maximul de concentraţie înregistrat pe
proba de gelatină menţinută în suc intestinal.

Analizând figura 16 putem confirma migraţia uniformă a acidului ascorbic în fluidele
biologice, fiind evident că fluidele biologice participante la digestie asigură cea mai bună
tranziţie a acidului ascorbic din gelatină.
3.10. Studii privind stabilizarea alcoolului polivinilic cu extract din pieliţă de
struguri
Luând în considerare diversele aplicaţii ale acidului polivinilic în calitate de biomaterial,
în cadrul aceastei teze de doctorat a fost cuprins şi studiul acestui polimer. Probele preparate,
alcool polivinilic la care s-au adăudat diferite concentrații de resveratrol și apă au fost menținute
menţinute 7 zile la 36,6 °C pentru a investiga posibilele schimbări intervenite asupra materialului
la această temperatură. Apoi, au fost analizate prin spectroscopie FT-IR.

La prima vedere, se observă clar că există diferenţe semnificative între spectrele acestor
probe. Transmitanţa probelor cu conţinut mai mare de extract este mai redusă (în cazul probei de
alcool polivinilic cu 5 mL extract). Din analiza spectrelor FT-IR ale probelor de alcool
polivinilic tratate cu extract de Căpşunică, se pot observa mai multe peak-uri: 3320 cm-1

Pentru a aprecia diferenţele spectrale ale probelor de alcool polivinilic, spectrele
înregistrate au fost comparate folosind corelaţiile de tip Pearson și analiza componentelor
principale. Spectrele probelor se asemănă foarte mult mai ales în cazul probelor consecutive
(PVA_0 şi PVA_1mL_Ex; PVA_1mL_Ex şi PVA_3mL_Ex), excepţie făcând PVA_5mL_Ex
care se aseamănă mai mult cu primele 2 probe decât cu PVA_3mL_Ex. Analizând proba
PVA_5mL_Ex, este foarte clar faptul că aceasta nu are aceeaşi tendinţă ca celelalte probe, fiind
caracterizată de componenta 2, în timp ce celelalte probe sunt caracterizate de componenta 1.
Acest lucru se poate datora faptului că volumul de extract adăugat la probe este direct
proporţional cu creşterea transmitanţei în zona 848 – 364 cm-1
. În cazul unui exces de cantitate
adăugată, scade şi transmitanţa mai ales în domeniul 3700 – 2960 cm-1
. Domeniul respectiv este
datorat grupării OH, care la un exces de extract adăugat produce o absorbanţă mai intensă.
3.11. Studii privind stabilizarea chitosanului cu extract din pieliţă de struguri
După omogenizare, probele de chitosan, extract de căpșunică (diferite cantități) și apă au
fost menţinute 7 zile la 36,6 °C, apoi au fost investigate prin spectroscopie FT-IR.

Cantitatea de extract adăugată la probele de chitosan influenţează foarte mult spectrele.
După cum arată corelaţiile de tip Pearson, spectrele se aseamănă între ele mai ales din aproape în
aproape (> 0,899, p < 0,01). Cele mai importante corelaţii sunt înregistrate între Chitosan 0 şi
Chitosan 1mL Extract (0,996, p < 0,01). Cu alte probe, Chitosan 0 are corelaţii mai slabe, ceea ce
poate semnifica că probele cu o cantitate mai mică de extract au fost influenţate de temperatură,
dar şi faptul că odată cu creşterea concentraţiei de extract se modifică şi spectrul chitosanului.
3.12. Studii privind stabilizarea colagenului cu extract din pieliţă de struguri
În ceea ce privește investigarea colagenului, studiul s-a axat pe analizarea unor probe de
colagen stabilzate cu extract de Căpşunică prin spectroscopia FT-IR. Înainte de această
determinare, probele au fost menţinute timp de 7 zile la 36,6 °C.

În urma determinărilor FT-IR asupra probelor de colagen, au rezultat spectrele din figura
3.92. Au fost observate diferenţe majore între spectrele FT-IR ale probelor de colagen tratate cu

cantităţi diferite de extract de Căpşunică. Una din cele mai importante observaţii ar fi scăderea
transmitanţei în domeniile 3600 – 2700 cm-1
şi 1689 – 400 cm-1
cu creşterea cantităţii de extract
adăugat în probe (Figura 19).
O altă observaţie importantă ar fi că odată cu creşterea cantităţii de extract adăugate
creşte transmitanţa în domeniul 1820 – 1670 cm-1
(număr de undă caracteristic grupării
funcţionale carbonil C=O). Acest lucru poate semnifica faptul că procesele de oxidare sub
acţiunea temperaturii sunt inhibate de către compuşii fenolici din extract.
După aplicarea metodelor de analiză statistică a rezultat influența pe care o are cantitatea
de extract asupra transmitenței la numărul de undă 1800 cm-1
(0,924), caracteristică grupării
funcţionale carbonil C=O. Această influență conduce la o scădere aproape globală a
transmitanţei pe întreg domeniul spectral.
3.13. Studii privind stabilizarea hidroxilatului de colagen cu extract din pieliţă de
struguri

Prin aplicarea analizelor statistice am ajuns la concluzia că este posibil ca de la o anumită
concentraţie în probe, compuşii fenolici să aibă aceeaşi influenţă sau chiar să influenţeze negativ
stabilitatea probelor de hidrolizat de colagen.
3.14. Studii privind stabilizarea polistirenului cu extract din pieliţă de struguri
Polistirenul este un material cu durată foarte mare de degradare (peste 1000 de ani). De
multe ori în acesta sunt adăugaţi diverşi antioxidanţi şi stabilizatori artificiali care îngreunează
procesul de descompunere în mediu. Pentru a înlesni mecanismul de biodegradare, în acest
material s-a adăugat extract din pieliţă de struguri de soiul Căpşunică.

Pentru supunerea materialului la un stres fizic s-a folosit un mediu puternic oxidant pe
bază de HNO3 65 % şi H2O2 %.
Schimbările structurale au fost monitorizate prin spectroscopia FT-IR (Figura 21) și prin
cromatografia ionică.
De asemenea, proprietăţile de stabilizator al extractului de Căpşunică asupra
polistirenului au fost studiate prin calorimetria diferenţială (DSC).
Pentru a putea observa efectele stresului fizic (mediu oxidant puternic) asupra probelor de
polistiren, după separarea fazei lichide (în care se conţinea acidul azotic rezidual), probele au fost
analizate prin SEM.
Astfel, pentru a observa care dintre probele de polistiren este mai stabilă la mediul
oxidativ, se va analiza graficul privind corelaţiile de tip Pearson dintre proba martor (Polistiren
0) şi celelalte probe. Rezultatele acestei prelucrări au fost expuse în grafic. Se poate observa că
cea mai mare stabilitate la agenţi oxidanţi este dată probele intermediare de polistiren, care au
avut un adaos de aproximativ 3 mL extract de Căpşunică. Un adaos prea mic (ex: 0,5 mL) sau
prea mare (5 mL) afectează stabilitatea polistirenului.
Determinările de cromatografia ionică s-au efectuat nu direct asupra probelor, ci asupra
lichidului rămas în urma reacţiei.
După cum am menţionat anterior, în calitate de agenţi oxidanţi, a fost folosit un amestec
de HNO3 65% + H2O2 30 %.
Pentru a putea investiga modul de interacţiune a amestecului oxidant asupra probelor de
polistiren, s-a analizat cantitatea de acid azotic rezidual, care nu a reacţionat cu proba,
observându-se astfel cum cantitatea de extract de Căpşunică măreşte sau scade
biodegradabilitatea acestui material polimeric. Extractul de Căpşunică, având proprietăţi
antioxidante mai slabe faţă de alţi antioxidanţi sintetici, a cedat mai uşor în condiţii puternice
oxidante.
Acest lucru poate duce la scăderea timpului de degradare a polistirenului în mediul
înconjurător.
Probele au fost preparate, prin diluţii în aşa fel încât să se încadreze în curba de calibrare
a azotatului folosită pentru cuantificarea rezultatelor în cadrul tehnicii de cromatografie ionică.
S-a putut observa că odată cu creşterea concentraţiei de extract de Căpşunică, cantitatea de acid
azotic rezidual scade progresiv.
Este posibil ca acidul azotic să reacţioneze cu polistirenul, reacţionând cu nucleele
benzenice ale acestuia, dar nu este exclusă probabilitatea reacţiilor paralele şi cu extractul, care
este bogat în compuşi fenolici.
Astfel, cantitatea de acid azotic reziduală, rămasă în soluţie este descrisă de o ecuaţie
exponenţială privind reacţia cu materialul polimeric îmbogăţit (y = 15676e-0.644x, R² = 0,9063),
având un R2 destul de mare (> 0,9). Cantitatea de azotat rezidual scade exponenţial de la 16 g/L
la < 2 g/L odată cu creşterea concentraţiei de extract din probe.
În concluzie, prin adăugarea de extract de Căpşunică materialul polimeric devine mai
vulnerabil la acţiunea factorilor de stres fizico-chimici, crescându-i în aşa fel biodegradabilitatea
în mediu.
După separarea fazei lichide, probele de polistiren au fost analizate prin SEM.
Efectele negative ale agenţilor oxidanţi puternici afectează structura materialului.
Spre exemplu, în figura 22 este reprezentat polistirenul pur. Acesta are un aspect
compact, fără fisuri. Deşi proba a fost supusă acţiunii agenţilor oxidanţi puternici, aceasta a fost
afectată doar la suprafaţă, și într-o măsură foarte mică.

În figura 23, este observată proba PolistirenM 3, în care au fost adăugaţi 3 mL extract de
Căpşunică, după care a fost supusă condiţiilor oxidante. Proba este afectată în mod vizibil, iar pe
suprafața acesteia există fisuri.

În cazul probei de PolistirenM 5 (Figura 24), în care a fost adăugați 5 mL de extract de
Căpşunică, se pot observa, de asemenea, consecinţele degradării de către agenţii oxidanţi
puternici. Fisurile sunt mult mai evidente, iar formaţiunile libere de material sunt într-o cantitate
mai mare pe suprafaţa materialului.

Putem concluziona că după o anumită concentraţie extractul nu numai că nu stabilizează
materialul polimeric, dar este și sursa producerii unor reacţii nedorite, care afectează stabilitatea
materialului. Suprafaţa materialului este mult mai denivelată faţă de PolistirenM 3, iar defectele
sunt mult mai vizibile.
Pentru determinările DSC au fost folosite probe de polistiren îmbogăţit cu extract de
Căpşunică, dar fără a fi supus la condiţii puternic oxidante.
Testarea stabilității prin calorimetrie diferenţială a avut drept scop investigarea
comportamentului probelor de polistiren la temperatură.
Din termogramele obținute se observă că probele au un comportament foarte diferit
(datorită prezenţei extractului de Căpşunică); acesta măreşte cu aproape 20 °C temperatura de
începere a proceselor de oxidare (în cazul probei de PolistirenM 1,5).
Odată cu creşterea concentraţiei de extract de Căpşunică în probe, începe să scadă
temperatura de începere a proceselor de oxidare, ajungând pentru proba PolistirenM 5 chiar mai
mică decât în cazul probei martor (figura 25).
Maximul temperaturii de începere a proceselor de oxidare (220 °C) este la probele
PolistirenM 1 şi PolistirenM 1,5, deşi cu creşterea cantităţii de extract temperatura această scade
lent până la 210 °C (pentru proba PolistirenM 4), după care scade brusc la 191 °C, în cazul
probei PolistirenM 5.

CONCLUZII
Teza cu titlul: Materiale biodegradabile cu antioxidanți naturali pentru protecția
mediului si aplicații biomedicale a avut drept scop cercetarea şi obţinerea unor materiale
biodegradabile stabilizate cu antioxidanţi naturali.
Realizarea acestei lucrări a avut la bază atât un suport teoretic, cât şi unul experimental.
Toate observaţiile identificate experimental au fost explicate folosind tehnici matematice şi
statistice de interpretare. Astfel, a fost posibilă prelucrarea unui volum mare de date, obţinând
rezultate de mare interes.
Resveratrolul este antioxidantul de interes din prezenta lucrare, acesta fiind cunoscut
datorită efectelor biomedicale şi capacităţii sale de apărare a organismului împotriva bolilor de
inimă coronariene şi întârzierea proliferării celulelor tumorale. Resveratrolul din struguri este
conținut aproape în întregime în pieliţa de struguri, iar nivelul maxim de concentraţie este atins
înainte ca strugurii să ajungă la maturitate.
Au fost obţinute extracte din pieliţă de struguri, aparţinând mai multor soiuri existente pe
piață şi anume: Căpşunică, Tămâioasă neagră, Hamburg, Moldova, Coarne Moldoveneşti. Ca
solvent de extracție a fost folosit etanolul.
Pentru a putea integra resveratrolul în structura diverselor biomateriale, au fost necesare
studii privind modul de introducere a acestuia în matricea materialelor biodegradabile. Cel mai
important aspect este stabilitatea resveratrolului la temperatură, pentru că majoritatatea
materialelor polimerice sunt prelucrate prin tratament termic.
Antioxidanţii naturali (şi extractele cu conţinut ridicat al acestora) au fost caracterizaţi
prin spectrofotometria UV-Vis, folosind metoda Folin-Ciocalteu (la aproximativ 760 nm), dar şi
prin metoda determinării conţinutului de compuşi fenolici la 280 nm. Au fost efectuate studii
privind stabilitatea resveratrolului (antioxidant natural) la temperatură ridicată cu ajutorul
spectrofotometriei UV-Vis şi spectroscopiei FT-IR.
Extractele din pieliţă de struguri au fost analizate prin spectrofotometria UV-Vis, folosind
metoda Folin-Ciocalteu, dar şi prin spectroscopia FT-IR şi calorimetria diferenţială (DSC).
Gelatina a fost unul din principalele materiale biodegradabile investigate, căreia i-a fost
studiat comportamentul termic, dar şi comportamentul în diferite fluide biologice (suc gastic, suc
intestinal, plasmă sanguină). Studiile pe gelatină au fost efectuate folosind spectrofotometria
UV-Vis şi spectroscopia FT-IR.
Pentru studiul polistirenului stabilizat cu material polifenolic (extract din pieliţă de
struguri) au fost folosite calorimetria diferenţială (DSC), spectroscopia FT-IR, cromatografia
ionică (IC) și microscopie electronică de baleiaj (SEM).
Prezenta lucrare cuprinde şi studii privind:
- stabilizarea colagenului cu antioxidanţi naturali;
- stabilizarea chitosanului cu antioxidanţi naturali;
- stabilizarea hidrolizatului de calogen cu antioxidanţi naturali;
- stabilizarea alcoolului polivinilic cu antioxidanţi naturali.
Plecând de la obiectivul principal al acestei teze de doctorat, cercetarea a avut ca scop
producerea de materiale biodegradabile cu antioxidanţi naturali pentru aplicaţii biomedicale şi de
mediu.
Prin coroborarea tuturor datelor obţinute şi prin comparaţia cu literatura de specialitate, se
poate menţiona că prezenta teză are un grad înalt de originalitate, combinând nu numai metode
de investigaţie analitică, dar şi metode statistice de interpretare corectă şi obiectivă a datelor
obţinute.
Cercetările efectuate pe diverşi antioxidanţi şi compoziţii bogate în antioxidanţi au dus la
noi metode de cercetare a stabilităţii acestora.
Prezenta teză de doctorat are un grad înalt de noutate şi originalitate, fiind orientată către
un subiect foarte important de utilizare a antioxidanţilor naturali în stabilizarea materialelor
biodegradabile.
Diseminarea rezultatelor obţinute s-a făcut prin publicarea de articole în reviste naţionale
şi internaţionale de prestigiu şi cu participări la conferinţe internaţionale în vederea prezentării
rezultatelor cercetării.

THE THESIS’ STRUCTURE
This doctoral thesis titled "Biodegradable Materials with Natural Antioxidants for
Environmental and Biomedical Applications" consists of 4 chapters, which summarizes a
comprehensive study of current scientific literature, but also an experimental side, featuring a
wealth of data on the stabilisation of the biodegradable materials with natural antioxidants.
CHAPTER 1 refers to the epistemological positioning of the research, including the
objective and the research issues, assumptions, analitical methods, statistical research and the
structure of the thesis.
CHAPTER 2 includes theoretical part about stabilising biodegradable materials with
natural antioxidants, explaining the properties of polymeric materials studied, but also natural
antioxidants . This chapter provides a brief overview of the current problems on the research of
biodegradability materials used and their prospects.
CHAPTER 3 may be called surely the most valuable and important part of this study. In
this part of the thesis, there were characterised materials used, using analytical techniques with
which the investigations on the material had been made, statistical methods used for the
interpretation of the results.
Natural Antioxidants (and extracts with high content of them) have been characterized by
by UV-VIS spectrophotometry, using Folin-Ciocalteu method (at about 760 nm), but also by the
method of determining the content of phenolic compounds at 280 nm. Studies have been
regarding the stability of resveratrolului (natural anti-oxidant) at high temperature using UV-VIS
spectrofotometry and FT-IR.spectroscopy
The extracts from grape skin were analyzed by by UV-VIS spectrophotometry using the
Folin-Ciocalteu method, but also through FT-IR Spectroscopy and differential calorimetry
(DSC).
Jelly studies have been carried out by using UV-VIS spectrophotometry and FT-IR
Spectroscopy.
For polystyrene study stabilised with polifenolic material (extract of grape skin)
were used differential calorimetria (DSC), FT-IR spectroscopy, electron microscopy with
sweeping (SIG) and ion chromatography (IC). Studies related to chitosan, polyvinyl
alcohol, collagen and hidroxilat of calogen have been carried out by FT-IR spectroscopy.
CHAPTER 4 reaches the main conclusions on this scientific work, underlying
element’s originality and the novelty of this thesis.
CHAPTER 5 includes bibliographical references used in the framework of the
doctoral thesis.
CHAPTER 1. THE PROCEDURE OF THE EPISTEMOLOGICAL
REASERCH
Taking into consideration the deplition of the petroleum reserves of the planet, current
investigations are aimed at renewable use of raw materials and the generation of biodegradable
materials. From this point of view, vegetable raw materials and particularly biopolymers, possess
properties of great interest in the industry of plastic materials such as: biodegradability,
biocompatibility, selective permeability and the unchangeable physico-mechanical propreties.
The ability to obtain new materials, with enhanced properties, by mixing two or
more polymers represent a better and practical solution than the design and synthesis of
new polymers, economically speaking. Over the past few decades, scientific research has
been directed toward developing a new class of polymers, the biomaterials.
One of the most discussed natural antioxidants remains resveratrol. Its properties
have been very extensively studied by researchers, who have found especially beneficial
effects on health (1; 2).
In order to integrate the structure of the resveratrolin various biomaterials, studies
on how to enter the matrix in its biodegradable materials were necessary to be made. The
most important aspect is the stability of the resveratrol to temperature, because most
polymeric materials are processed by heat treatment.
There are other ways to integrate the antioxidants in the biodegradable materials
structure. On the basis of the polymer solubility in a given solvent it is possible the
introduction of certain antioxidants in the polymer. For example, the jelly can be enriched
with anti-oxidant by dissolving it in the water, or polystyrene – by disolving it in
chloroform.
Nowadays, degradable polymers are of special interest, such as for example:
chitosan, collagen, hydroxylated collagen and polyvinyl alcohol. They are for a long time
used in biomedical applications and have high degree of biocompatibility. Stabilizing these
polymers with natural antioxidants, can increase the life of these materials, but it also
decreases the risk of poisoning due to the replacement of the artificial indices with the
naturals.
Generally speaking, the interaction between any polymeric material with natural
antioxidants brings a considerable advantage on the degradability for the environment. In
the medical field, the use of the natural antioxidants in different biodegradable materials
would produce many positive effects.
The main objective of this thesis has been to produce biodegradable materials stabilized
through natural antioxidants with applications in medicine and for the protection of the
environment. The problematic research emerged due to the toxicity produced by the artificial
antioxidants on the environment. Also, in the case of poorly degradable polymeric material the
replacement of the artificial indices with the natural ones would decrease the duration of their
degradation and would increase the chances to biodegradability.
CHAPTER 2. THE DEGRADABILITY AND BIODEGRADABILITY OF
THE POLYMERIC MATERIAL UNDER THE ENVIRONMENTAL
CONDITIONS
Synthetic polymers caused a great enthusiasm from a technological point of view, so that
in the mid-'50S commissioning of new production facilities and the research of obtaining new
polymeric structures for civil applications, industrial and military were in full momentum.
The awareness of the problems that these materials can create on the environment
appeared much later, when their accumulation and persistence to landfill or in ambient
environment became obvious, when effects on flora, fauna and the quality of the soil and water
have been proven and understood to a greater extent. In addition, the oil crisis in the late ’70s
revealed that ensuring raw materials resources for the continuation and development of
production of polymeric material could be itself a problem in the future (3).
In accordance with the conditions for using materials derived from renewable resources,
which would contribute to maintaining virtually constant concentration of CO2 in the
atmosphere, both waste disposal of plastic materials by combustion and by degradation in the
environment can win in efficiency, due to the use of sources of raw materials alternative to those
fossil fuels. An intermediate study represent the use of traditional polymeric material with
properties that are controlled by means of additives which have a lower impact on the
environment and human health. Thus, it is well known the use of the phenolic indices for the
stabilization of poliolefines during processing and to ensure a certain durability during use. It is
well known that phenolic antioxidants can show an important bacteric activity and even a certain
toxicity to animals and humans. These additives can migrate in foodstuffs intended to come into
contact with the polymeric packaging materials, but also in the ground or in the water under the
conditions of polimeric material degradation into the environment. (4)
A solution for the reduction of risks related to the use of additives (antioxidants,
stabilizers) on human health and the environment is the use of natural products or synthesis
products with a similar structure to those natural ones. (4)).
Concerns about finding some polymeric structures that are able to suffer degradation in
active surrounding environments (Figure 1), under the action of the micro-organisms, started in
the 1980s. Biodegradable plastic materials have opened a way for new strategies in the
management of waste because such materials are designed to degrade over environmental
conditions or in industrial installations of biological treatment of waste (5).

Evaluating susceptibility to biodegradation, and setting up mechanisms of reaction, of the
factors of influence and their proportion, thereof provides both an opportunity to govern the
biodegradation process, as well as the synthesis of materials whose waste to be easily integrated
in natural ecosystems. (7)
After exposure to conditions defined by international standards (ASTM - American
Standard Testing Methods or ISO - International Standards Organization), samples are analyzed
from the point of view of physical properties, chemical or biological. The results obtained should
be compared with the values obtained from homonymous control samples.
Thus, there have set biodegradation tests in the soil or in activated sludge under
controlled conditions of temperature, humidity, and ventilation. In general, the testing of
biodegradability can be carried out according to the protocol in figure 2. (8)

Nowadays, they are making efforts to transform existing nondegradable polymers in
degradable polymers, photodegradable or hidrolizabble by changing chemical or through the
inclusion of additives (for example, senzitives and biopolymers) and through the development of
methods which make it possible to use the native degradable polymers (e.g. Poles (beta-
hidroxibutirat)). (9; 10]
Appropriate use of degradable polymers in different applications depends on the
knowledge of interaction between these materials and the environment. This interaction is
controlled, among other things, by the type and quantity of the small molecules that could
penetrate any material or emits from it by diffusion and migration (10). Industrial production of
many of the degradable polymers is still very expensive for applications such as single use items

(movies packing, diapers, disposable cutlery), but in a range of biomedical applications, the cost
has a minor importance.
Nowadays, antioxidants are used on a large scale in the food industry to prevent or delay
lipid oxidation. The oxidation control is essential in the management of developments in
biological systems in their complexity, in particular in the case of foodstuff because degradation
may affect the safety of food (Figure 3). (11)
There are several criteria that must be taken into consideration when choosing appropriate
antioxidants to be included in food products, such as: the cost price, its toxicity, the efficacy of
the small concentrations, stability, and the ability of the processing, the color, flavor, and the
smell (12)).

The role of phenolic compounds in the body is generally for protection against certain
diseases due to possible interactions with enzymes and their antioxidant properties. Specifically,
flavonoids are assigned to a wide variety of properties, namely: antitumor, antiinflammatory,
painkillers, allergenic, antispasmodic, antibacterial, hepatoprotective (13)). They limit the
oxidation of the fat from blood and contribute to the fight against atheromatous plates. They also
have anxiolitic properties protecting arteries against atherosclerosis and trombosis. (14)
CHAPTER 3. EXPERIMENTAL PART
3.1. Preparing the samples
In this thesis I used samples from the grapes, belonging to many types such as:
Căpşunică, Tămâioasă neagră, Hamburg, Moldova, Coarne Moldoveneşti ( marked with P1, P2,
P3, P4, P5)
I have chosen ethanol as extraction solvent because it is not toxic, as shown in numerous
studies and with the ability to isolate a wider spectrum of extraction and polar compounds in the
sample. Grape skins have been removed manually, and then weighed (~ 100 g skin) and then
entered in 50 ml of ethanol 96 %. After mixing, the samples were kept in the dark for 48 hours.
The extracts were then obtained by centrifugation at 4000 rpm for 20 minutes, they have been
filtered through the filters with diam. 0,45μm and stored in the dark.
Preparation of aqueous solutions of trans-resveratrol
Trans-resveratrol (pure analitical substance 98%) dilute with 200 ml of H2O, resulting in a
solution of 31.25mg/l (137mm). Of this solution has been obtained aqueous solution of trans-
resveratrol with concentrations of 5, 10, 15,625.25 and 31.25mg/L.

To determine the concentration of resveratrol of the studied extracts, it have been
performed 3 calibration curves at 3 different wave lengths (252.286, 304 nm) (15) using pure
analytical resveratrol (Merck) and using an Analityk Jena- Specord 250 Plus spectrophotometer.
The concentration of the resveratrol- equivalentfrom the grapes samples had beea
determined using the calibration curve of the resveratrol at 304 nm (Figura 4).

Preliminary data of the UV-VIS spectroscopy showed a higher content of resveratrol
equivalent in samples P1 and P4 and lower in P2 and P3.
3.2. Determination of total phenol index in the extracts from grapes
The polyphenols total index has been determined by measuring the absorbance at 280 nm
using vats of optical path of 10 mm and making dilution of 1:100 AM (Figure 5) (16, 17). The
results obtained are in good correlation with the concentration of equivalent-resveratrol from
grape extracts. Thus, the content of polyphenols is higher in samples P1 and P4, and lowest in
samples P2 and P3. A very interesting phenomenon can be observed between the polyphenols
total index and the concentration of equivalent-resveratrol extracts (Figure 6). An almost linear
relationship between them can be observed. Of course, mathematical relationship between these
two indices (R2 0,8576 ) may be a result of rapprochement between wavelengths at which the
measurements were carried out (280 and 304 nm).

3.3. Determinările FT-IR efectuate pe extractele de struguri şi resveratrol

Using this technique there have been identified the functional groups in the composition
of grape extracts (Figure 7) and have been obtained more data concerning the extracts analyzed.
For recording of the spectrum the use of an aqueous solution of trans-resveratrol has been made,
having the concentration of 15,625 mg/L.

From the analysis of the obtained graphs it can be observed the peaks 1240 cm-1.600 -
800 cm 1.3200 - 3400 cm-1, drops that characterise functional groups present in aromatic
compounds. As known, in grapes the most encountered aromatic compounds are the
polyphenols. It can be concluded analysing the acquired information that FT-IR spectroscopy
may be useful in detecting polyfenols (Figure 7-8).
3.4. Differential calorimetry determinations, carried out on the grape extracts
Measurements related to differential calorimetry (DSC) have been carried out on 2
extracts of grapes, namely Căpșunică (labeled Q1, Figure 9) and Hamburg (labeled Q2, Figure
10), which have passed through a special preparation (dehydration at 400 °C for long periods).

It was noted that processes which are likely to influence the concentration of polyphenols
begin after temperature of 170oC, as we are talking about oxidation, and after 190
oC begins the
processes of decay. It should be noted that in the extract Q1 these processes begin to higher
temperatures (oxidation at 190oC, decomposition at 200
oC).
3.5. The characterization of the antioxidant activity of the grapes extracts by the
Folin-Ciocalteu method
The use of Folin-Ciocalteu index has been made to characterize antioxidant activity of
the extracts of grapes, measurements being made at 4 wavelengths: 725.750, 760, and 765 nm.
Pearson type correlations from the concentration values of grapes extracts obtained from the
calibration curve to 4 wavelengths (725.750, 760, and 765 nm) showed very large similarities
(1,000, p < 0.01 ) (Table 1). This statistical analysis couldn’thighlight the differentiation between
the concentrations of gallic acid from the samples.

There were applied probability parameters of the true concentration of phenolic
compounds in the analyzed extracts. By analysing main components, followed by the Varimax
method of rotation with Kaiser normalization, it were obtained 2 main components (Figure 11):
- Component 1, was characterised by the values of grape extracts concentrations,
which showed relatively high standard deviation;
- component 2, which is characterized by the values of grape extracts
concentrations, which showed relatively small standard deviation.

3.6. Investigations regarding the thermal stability of the resveratrol
Different samples of aqueous solution of trans-resveratrol (19 mg/L resveratrol) have
been exposed to temperatures of 30.40, 50.60, 70.100 and 120oC for 30 minutes. In addition,
other samples having the same concentration have been exposed to temperatures of 30.40, 50.60,
70 oC. For determinations concerning the stability of the trans-resveratrol to temperature it have
been used a solution of resveratrol 19 mg/L. Investigations carried out have demonstated that
temperature has a significant effect on trans-resveratrol's absorbance at the wavelength 286 and
304 nm. Subtracting the absorbance demonstrates that at high temperatures, resveratrol shall be
destroyed. The conversion of the trans-resveratrol in cis-resveratrol shall not be admissible due
to the fact that the cis-resveratrolul present in the solution seems to be degraded.

3.7. Wavelength Study for the Folin-Ciocalteu test

The wavelengths at which the Folin-Ciocalteu index determinations have been are: 725,
750, 760 şi 765 nm (Figura 13). The calibration standards were prepared from a stock solution of
5 g/L Gallic acid.
By analysing the spectra of Gallic acid solutions and applying the 3 types of statistical
analysis (linear regression analysis, main components, Pearson type correlations) it can be
concluded that the 725 nm wavelength is the most appropriate. Thus, at a 725 nm wavelength
there is the strongest correlation between the absorbance and concentration, and the maximum
absorption is around 765 nm.
3.8. Studies on system interactions jelly-resveratrol-water
To investigate system gelatine-resveratrol-water behavior, five samples with the same
quantity of gelatine have been prepared, but with various concentrations of resveratrol. Prepared
samples have been subjected to a heat treatment at temperatures of 40.60, 80 °C, and then they
were analyzed by FT-IR spectrometry.
It was noted that the dehydration processes for the gelatine-resveratrol-water system are
not affected by the amount of resveratrol added, and samples behave alike at a temperature of 40
°C ( >= 0,897, p < 0.01 ). By applying these Pearson correlations it was noted that rasveratrol has
no influence or has an insignifican tinfluence on the loss of mass of the sample.
From the FT-IR spectres analysis was noted that there are differences between samples in
the 8000 - 400 cm-1 field. This may be due to proteins of the gelatine which hydrolyse in a
different way, since it has been demonstrated by statistical analysis of the main components that
at this temperature the resveratrol has an insignificant influence on gelatine.
Regarding the impact that the time has on gelatine samples stabilized with resveratrol and
kept at 60 °C, should be noted that it would have a smaller impact ( >= 0,601) compared with the
samples that have been maintained at a temperature of 40 °C. The explanation might be in the
fact that higher temperature accelerates loss of mass (especially loss of water) in a shorter time.
Shall be observed very close correlations between mass losses of the samples (1,000 ; p < 0.01),
thus demonstrating a balanced manner of transformation. There is an influence of the resveratrol
on the system observed after 48,06 h at a 60°C temperature, but characterised by an average
correlation (0,487 ).
Sample P5 (with the highest concentration of resveratrol) presented an intermediate
nature of the kinship between spectra. Distribution of samples in the graphic, from this point of
view, may be linked to, as it has been shown, to the hydrolysis processes and restore connections
for dehydration, which arose in a random manner.
With regard to the heat treatment at a temperature of 80 °C, the time influence is
described by strong enough correlations ( ≥ 0,705 ). There are established very strong correlation
between the values of loss masses ( ≥ 0,988, p < 0,05), showing that dehydration goes in the
same way for all the samples. As in the case of the other studies (at 40 and 60 °C), we have
noticed that the time has influence on samples, but not entirely.

From the spectres analysis and by the application of the statistical analysis methods it was
noted that the samples P3_40, P4_40 and P1_80 have higher transmittance in the 3300 - 1082
cm-1.field.
3.9.Studies regarding of the behavior of stabilised gelatine in different biological
environments
În prezenta lucrare a fost studiată interacţiunea dintre resveratrol şi gelatină, acid ascorbic
şi gelatină, extract struguri şi gelatină, extract rozmarin şi gelatină. Este foarte importantă
compatibilitatea dintre cele 2 componente, dar şi comportamentul fiecărui sistem in interacție cu
diverse fluide biologice: suc gastric, suc intestinal, plasmă sanguină.
In this work the interaction between resveratrol and gelatin, ascorbic acid and gelatin,
grape extract and gelatin, rosemary extract and gelatin has been studied. It is very important the
compatibility between the 2 components, but also the behavior of each system in interaction with
different biological fluids: gastric juice, intestinal juice, blood plasma.
The Folin-Ciocalteu test offered a very good determination of the amount of active
principles migrated from biological material fluids.
The extract of rosemary presented an high hydrophobicity in all biological fluids studied,
but it is possible that in natural biological environment things to go different.
It was noted, in all fluids analyzed, after an interval of 96 h, a migration of the active
principle (quantified as mg/L gallic acid equivalent), and the most significant values have been
observed in samples which have had direct contact with gastric juice and juice intestines. In
conclusion, the extract of rosemary can be used as a component in the composition of oily
principles or in environments where there is a need for the antioxidant not to migrate (Figure 14).
As for the quantity of skin grape extract (Căpşunică) added in the samples, it should be
noted that this is directly proportional to the concentration of Gallic acid equivalent of samples
that have been maintained in juice gastric and intestinal juice. Samples maintained in the blood
plasma have demonstrated an increase of the phenolic compounds in relation to time, but
uncorrelated to the quantity of grape extract added.

In intestinal juice took place the smoothest release of the polyfenol compounds. Samples
of gelatine maintained in gastric juice had an intermediary character, but, which combined with
the action of gastric juice could accelerate the migration of polyfenol compounds and the
hydrolysis of gelatine.
In figure 15 it is to be noted that in all biological fluids the diffusion takes place.
However in gastric juice and intestinal this result goes in a more accelerated rate.
Regarding the ascorbic acid it has been noted that its migration in gelatin is directly
proportional with the duration of the contact and with biological fluids, but also with the
concentration of ascorbic acid of the samples. The greatest amount of ascorbic acid concentration
is observed in gastric juice and in intestinal juice. The maximum concentration recorded on
gelatine maintained for 96 h in the blood plasma is 2 times lower than the maximum amount of
sample concentrations recorded on the gelatine maintained in intestinal juice.

By analysing figure 16 we can confirm uniform migration of the ascorbic acid in biological
fluids, clearly biological fluids participating in digestion provides the best transition of the
ascorbic acid from the gelatine.
3.10. Studies regarding the stabilization of polyvinyl alcohol with extract of grape
skin
Taking into account the various applications of polyvinyl acid as biomaterial, within the
framework of this doctoral thesis, it has been included the study of this polymer. Samples
preparations, polyvinyl alcohol to which it have been added different concentrations of
resveratrol and water, have been maintained 7 days to 36.6 °C to investigate possible changes on
material at this temperature. Then, they have been analyzed by FT-IR spectroscopy.

At first sight, it is noted that there are significant differences between spectra of the
samples. Transmittance of samples with a higher content of the extract is smaller (in the case of
polyvinyl alcohol with 5 mL extract). The analysis spectres FT-IR sample for polyvinyl alcohol
extract treated with Căpşunică, it may be observed several peaks: 3320 cm-1
(OH); 2910 cm-1
(CH); 1740 cm-1
; 1420 cm-1
; 1325 cm-1
; 1090 cm-1
(CO); 833 cm-1
; 472 cm-1
(Figura 17).
To apreciate differences in the polyvinyl alcohol sample spectral, the spectra recorded
were compared using Pearson correlations and main components analysis. Spectra of samples
closely resemble especially in the case of consecutive samples (PVA_0 şi PVA_1mL_Ex;
PVA_1mL_Ex şi PVA_3mL_Ex), with the exception of PVA_5mL_Ex which resembles more
with the first two samples than with PVA_3mL_Ex. Analysing PVA_5mL_Ex, it is very clear
the fact that this does not have the same trend as in the other samples, being characterised by
component 2, while the other samples are characterized by component 1.
This may be due to the fact that the volume of extract added to samples is directly
proportional to the higher transmittance in 848 - 364 cm-1
area. In the event of an excessive
amount of weight added, transmittance drops, particular in the 3700 - 2960 cm-1
field. That range
is due to OH, which in an added excess of extract, produces a more intense absorbance.
3.11. Studies regarding the stabilization of chitosan with extract of grape skin
After mixing, the samples of chitosan, extraction of căpșunică (different amounts) and
water, they have been maintained 7 days to 36,6 °C and then were investigated by FT-IR
spectroscopy.

The FT-IR spectra of pure chitosan shows peaks according to the wave numbers: 3385
cm-1
, 2880 cm-1
, 1654 cm-1
, 1600 cm-1
, 1081 cm-1
. In the case of FT-IR spectres obtained on
chitosan samples treated with extract of Căpşunică. I have noticed the main peaks at wave
numbers: 3330 cm-1
; 1573 cm-1
; 1410 cm-1
; 1016 cm-1
; 870 cm-1
; 646 cm-1
(Fig 18).
Cantitatea de extract adăugată la probele de chitosan influenţează foarte mult spectrele.
The quantity of extract added to the chitosan samples influences very much the spectra. As
Pearson type correlations show, the spectra resembles between them especially taken little by
little (> 0,899 , p < 0.01). The most important correlations are recorded between Chitosan 0 and
Chitosan 1mL Extract (0,996 , p < 0.01). With further samples, Chitosan 0 has weaker
correlations, which may mean that samples with a smaller amount of the extract has been
influenced by temperature, but also, the fact that with increasing concentration of the extract is
hereby modified the chitosan spectrum.
3.12. Studies regarding the stabilization of collagen with grape skin extract
In the case of invastigating the collagen, the study has focused on analysing samples of
collagen extract stabilized with Căpşunică by FT-IR spectroscopy. Prior to this determination,
the samples were kept for 7 days at 36.6 °C.

As a result of FT-IR determinations on samples of collagen, resulted the spectra in figure
3.92. There have been observed major differences between FT-IR spectra of samples of collagen
treated with different amounts of extract of Căpşunică. One of the most important observations
should be the decreased transmittance in the 3600 - 2700 cm-1 and 1689 - 400 cm-1 fields with
increasing the amount of the extract added to samples (Figure 19).
Another important thing would be that once the increase in the quantity of extract added,
transmittance increases in the 1820 - 1670 cm-1 field (number of wavelength characteristic to the
carbonyl functional group C=O). This may mean that oxidation processes under the action of the
temperature are inhibited by the phenolic compounds from the extract.
Following the application of the methods of statistical analysis, resulted the influence
over the quantity of extract on transmittance at wave number of 1800 cm-1(0,924 ), a
characteristic of the C=O functional grouping carbonyl. This influence leads to a reduction of
transmittance over the entire spectral range.
3.13. Studies regarding the stabilization of the hydroxylated collagen with skin
grape extract

The stabilization of the collagen extract was made with căpșunică extract. After mixing,
the samples were kept for 7 days at a temperature of 36,6 °C. Investigating the hydrolyzate
samples of collagen by FT-IR spectroscopy was also aimed at identifying structural changes on it
at a temperature of 36,6 °C for 7 days.
From the graphic it can be observed that once the increase in the quantity of extract
added, transmittance decreases at the wave numbers: 3600 – 2320 cm-1
and 1680 – 370 cm-1
(Fig
20).
In the case of hydrolyzate collagen samples there were observed peaks at the following
wave numbers: 3250 cm-1
; 2950 cm-1
; 1633 cm-1
; 1537 cm-1
; 1402 cm-1
; 1334 cm-1
; 1244 cm-1
;
1060 cm-1
; 522 cm-1
.
By applying statistical analysis I have come to the conclusion that it is possible that at a
certain concentration in the samples, phenolic compounds to have the same influence, or even
negatively influencing the stability of the hydrolyzate collagen samples.
3.14. Studies on the stabilization of polystyrene with grape skin extract
Polystyrene is a material with very long time of degradation (over 1,000 years). Many
times, are add various antioxidants and artificial stabilizers in it, which hinder the decay process
in the environment. To facilitate the mechanism of biodegradation, in this material was added
skin grape extract variety of Căpşunică.

To the submission of the material to a physical stress it has been used powerful oxidising
environment on the basis of HNO3 65 % şi H2O2 %.
Structural changes have been monitored by FT-IR spectroscopy (Figure 21) and by ion
chromatography.
Also, the properties of the Căpşunică extract stabiliser over the polystyrene have been
studied by differential calorimetry (DSC).
To be able to notice physical stress effects (an high oxidizing atmosphere) on samples of
polystyrene, after liquid phase separation (which contained residual nitric acid), samples were
analyzed by SEM.
So, in order to observe which of the polystyrene samples is more stable to oxidativ
environment, analysis will be regarding the graph of the Pearson type correlations between the
this sample (polystyrene 0) and all the other samples. The results of this process have been
exposed in the graphic. It can be seen that the highest stability against oxidizing agents is given
by the intermediate polystyrene samples that had an addition of approximately 3 ml of the
Căpşunică extract. An addition too low (ex: 0.5 mL) or too high (5 mL) affects the stability of
the polystyrene.
The ion chromatography determinations have not been carried out directly on samples,
but on the remaining liquid from the reaction.
As mentioned earlier, as oxidizing agents, it has been used a mixture of HNO3 65% +
H2O2 30 %.
To investigate the interaction of the oxidizing mixture on the polystyrene samples, it have
been analyzed the quantity of residual nitric acid, which did not react with the sample, thus
observing how the amount of the Căpşunică extract increases or decreases the biodegradability
of this polimeric material. The extract of Căpşunică, having weaker antioxidant properties than
other synthetic antioxidants, it gave up more easily into powerful oxidizing conditions.
This may lead, with time, to a decrease of degradation of the polystyrene in the
environment.
Samples have been prepared by dilution in such a way that it fits in the calibration curve
of nitrate used for the quantification of results in the context of ion chromatography. It could be
easily seen that with increasing concentration of the Căpşunică extract, the quantity of residual
nitric acid decreases gradually. It is possible that nitric acid to react with polystyrene, reacting
with the benzene cores of it, but it is not ruled out the possibility of parallel reactions with the
extract, which is rich in phenolic compounds.
Thus, the quantity of residual nitric acid remaining in the solution is described by an
exponential equation regarding the reaction of the sample with enriched polimeric material(y =
15676e-0.644x, R² = 0,9063), having a R2 large enough (> 0.9). The quantity of residual nitrate
decreases exponentially from 16 g/l < 2 g/L with increasing concentration of the extract from the
samples.
In conclusion, by the addition of the Căpşunică extract polimeric material becomes more
vulnerable to the action of physico-chemical stress factors, increasing their biodegradability in
the environment.
After the separation of the liquid phase, the polystyrene samples were analyzed by SEM.
Negative effects of strong oxidants agents affect structure of material.
For example, in Figure 22 is represented pure polystyrene. It has a compact appearance,
free of cracks. Although the sample has been subjected to strong oxidants agents, this was
affected only at the surface, and to a very small extent.

In figure 23, is observed the PolystyrenM 3 sample in which there have been add 3 mL
extract of Căpşunică, after which it has been subjected to oxidizing conditions. The sample is
clearly affected, and on its surface area there are cracks.

In the case of M 5 Polystyrene sample (Figure 24), in which it was added 5 ml of the
Căpşunică extract, it may also be noticed the consequences of degradation by strong oxidant
agents. The cracks are much more obvious and free groups of material are in a larger quantity on
the surface of the material.

It can be concluded that after a certain concentration the extract not only that it doesn’t
regulate the polimeric material, but it is also the source of undesirable reactions, which affects
the stability of the material. Material surface is much more uneven than that of the M 3
polystyrene, and faults are more visible.
For DSC determinations there have been used polystyrene sample extract enriched with
Căpşunică, but without being subject to the powerful oxidizing conditions.
Stability testing by differential calorimetry was also aimed at investigating behavior of
the polystyrene samples to temperature.
From thermograms obtained shall be noted that the samples have a very different
behavior (due to the presence of the Căpşunică extract); it increases by almost 20 °C the starting
temperature of the processes of oxidation (in the case of Polystyrene sample M 1.5 ). With the
increase in the concentration of the Căpşunică extract in the samples, the starting temperature of
the oxidation processes begins to drop, reaching for the M 5 sample even less than in the case of
the blank sample (Figure 25).
Maximum temperature of the start of the oxidation processes (220 °C) is to test M 1 and
M 1.75 polystyrene samples, although with the increase in the quantity of extract, the
temperature drops slowly up to 210 °C (for polystyrene sample M 4), then drops to 191°C, in the
case of M 5polystyrene sample.

CONCLUSIONS
The thesis with the title: Biodegradable Materials with Natural Antioxidants for
Environmental and Biomedical Applications aimed to research and to get some biodegradable
materials stabilized with natural antioxidants.
Carrying out this work was based on both a theoretical support, and an experimental one.
All experimental observations identified have been explained using mathematical and statistical
techniques of interpretation. So, it has been possible processing of a large amount of data,
obtaining results of great interest.
Resveratrol is the antioxidant of the most interest in this work, being known because of
its biomedical effects and its ability to defend the body against coronary heart diseases and delay
of tumor cells proliferation. The Resveratrol from grapes is contained almost entirely in the skin
grape, and the maximum concentration is reached before the grapes reach maturity.
Extracts were obtained from skin grapes, belonging to more varieties available on the
market, namely: Căpşunică, Tămâioasă neagră, Hamburg, Moldova, Coarne Moldoveneşti.
Ethanol was used as extraction solvent.
To be able to integrate the resveratrol in the structure of various biomaterials, studies
were needed on how to introduce it into biodegradable materials matrix. The most important
aspect is the stability of resveratrol to temperature, because most polymeric material are
processed by heat treatment.
Natural antioxidants (and extracts with high content of them) were characterized by UV-
Vis spectrophotometry using Folin-Ciocalteu method (approximately 760 nm), but also by the
method of determining the content of phenolic compounds at 280 nm. Studies were conducted on
the stability of Resveratrol (natural antioxidant) at high temperature using UV-Vis
spectrophotometry and FT-IR spectroscopy.
The extracts from grape skin were analyzed by UV-VIS spectrophotometry using the
Folin-Ciocalteu method, but also through FT-IR spectroscopy and differential calorimetry
(DSC).
Gelatin has been one of the main biodegradable materials investigated, to which it has
been studied its thermal behavior, but also its behavior in various biological fluids (gastric juice,
juice intestinal blood plasma). Gelatin studies have been carried out by spectrophotometry using
UV-VIS and FT-IR spectroscopy.
For the study of the stabilized polystyrene with polifenolic material (extract of grape
skin) were used differential calorimetry (DSC), FT-IR spectroscopy, ion chromatography (IC)
and electronic microscopy sweep (SEM).
This work also includes studies on:
- stabilizing collagen with natural antioxidants;
- stabilizing chiosant with natural antioxidants;
- stabilizing hydrolyzed collagen with natural antioxidants;
- stabilizing polyvinyl alcohol with natural antioxidants.
Starting from the main objective of this doctoral theses, the research aimed at the
production of biodegradable materials with natural antioxidants for biomedical and
environmental applications.
By collating data obtained through comparison with the specialized literature, it can be
noted that this thesis has a high degree of originality, combining not only analytical investigation
methods, but also statistical methods of correct and objective interpretation of the obtained data.
Researches on various antioxidants and antioxidant-rich compositions, have led to new
methods of research of their stability. This PhD thesis has a high degree of novelty and
originality, being geared to a very important subject for the use of natural antioxidants in the
stabilisation of the biodegradable materials.
The dissemination of the obtained results have been made through publication of articles
in national and international prestige magazines and with participations in international
conferences with the purpose of presenting the research results.
